Yan Barthelemy beat previously undefeated Roberto Benitez (6-1-0, 3 KOs) by 4th round TKO in a super bantamweight fight on Friday, April 2, 2010 at the Mohegan Sun Casino in Uncasville, Connecticut. The fight was stopped at 1:16 of round four due to a cut over Benitez's left eye.

Yan Barthelemy from Havana, Cuba defected to Columbia in December 2006 along with Cuban national boxing teammates Odlanier Solis also from Havana and Yuriorkis Gamboa from Guantanamo. The trio had been training in Venezuela with the Cuban team when they defected.

Barthelemy, a southpaw and 2004 Olympic gold medalist signed with promoter Ahmet Oner of Germany's Arena Box Promotions along with Solis and Gamboa. Golden Boy Promotions has also signed co-promotional deals with Barthelemy.

Barthelemy made his pro debut on April 27, 2007 at the Arena Gym in Hamburg, Germany against Russian Ravil Mukhamadiarov winning a 4-round UD. In his second pro fight on Saturday, June 16, 2007 at the Sport Salonu in Ankara, Turkey, Barthelemy beat Ukrainian Gennadiy Delisandru by PTS.

On Tuesday, October 30, 2007 at the Hard Rock in Hollywood, Florida, Barthelemy fought his first professional fight in the US beating Kevin "Bobo" Hudgins (5-12-0, 2 KOs) by a 4-round unanimous decision. The scores were 60-53, 60-53 and 60-53.

Cuban defector Barthelemy beat Jose Garcia Bernal (27-11-1, 18 KOs) by 6-round unanimous decision on Saturday, April 12, 2008 at St Pete Times Forum in Tampa, Florida. Scores were 60-53, 60-53 and 60-53. Bernal was down in round five.

Barthelemy lost to journeyman Ernie Marquez (7-4-0, 3 KOs) of Colorado by 6-round unanimous decision on Friday, August 1, 2008 at the Desert Diamond Casino in Tucson, Arizona. It was Barthelemy's first professional loss. The judges scores were 55-58, 55-56 and 55-56. Had Barthelemy not lost a point for excessive holding in round six the fight would have been ruled a majority draw.

on Friday, May 15, 2009 at the Mahi Temple Shrine Auditorium in Miami, Florida, Yan Barthelemy beat Alexander Espinoza (8-1-0, 7 KOs) by 10-round unanimous decision. In addition to his victory Barthelemy picked up the vacant WBC Latino Bantamweight Title. Barthelemy was coming off a 3rd round TKO win over Pedro Rincon Miranda (30-15-2, 25 KOs) on Friday, March 6, 2009 also at the Mahi Shrine Temple.

Barthelemy lost to Jorge Diaz (10-0-0, 6 KOs) by 6th round TKO in a super bantamweight fight on Saturday, October 10, 2009 at Madison Square Garden in New York. In a competitive fight Diaz trained by the late Arturo Gatti's assistant trainer Mikey Skowronski hit Barthelemy with a left hook to the chin at 1:07 of round six sending Barthelemy face first on to the canvas for the count and then some. Diaz vs Barthelemy took place on the Latin Fury 12 card promoted by Bob Arum of Top Rank, Ahmet Oener of Arena Box-Promotion and Ivan Rivera of Puerto Rico Best Boxing which co-featured Juan Manuel Lopez vs Rogers Mtagwa and Yuriorkis Gamboa vs Whyber Garcia.
